I resealed my bathtub and shower this weekend with silicone where the tile meets the tub. Now I noticed a questionable spot. Can I go over the spot in question again with silicone or do I have to strip all the new silicone sealant out and start over?

I'd like to say yes. I've done it and that's what the warranty guy did as well. Just make sure it's clean before you add it.

On a related note, how do you clean mold that's grown in the grout? =/ I've tried a 50/50 mix of bleach and I soaked a towel and left it on overnight and it didn't go away. Don't want to tear it out since it's a new home, but if that's what I got to do.....

Yes you can if the area is clean. Also you may need to go with an acid wash for the grout bro.
