In this specific video she is clearly stating her frustration that the first woman senator in SC should have been elected not just given the position. She sees it as a historical thing, is not that she is against a woman being a senator.

Pretty much everybody figured Graham was gay. While I'm guessing Graham went to enormous lengths to make sure it was never publicly known, I don't think it's what motivated his Trump flip. I think it's much simpler than that. Graham was just a craven opportunist. As soon as it became politically expedient to back Trump, he did.
