god damn I laughed so hard when I read the OP. I don't think its rude as long as your mother/grandmother found it to be humorous. It all depends on the situation and the people you are with. For some people, the exact thing that Pun posted would be rude...but with others, it's hilarious.

I tell you I gotta plead ingnorance on this thing because if anyone had said anything to me at all when I first started playing that that sort of thing was frouned upon, you know, cause I've been around a lot of grandmothers and I tell you peope do that all the time
When my older brother was 17 and a senior in high school, my Mom asked him if he was sexually active, then without waiting for an answer gave him an envelope full of condoms. I still wouldn't feel comfortable spelling, then explaining, queefs to her in a game of Scrabble.
The world must be about to spin off its axis and crash into the sun, because I'm about to agree with bigtexxx. I can't even imagine playing the word queef with my mother, much less grandmother.
Well, I'm gonna pull a 180. The more I think about it, the more I realize that one of crappiest things about being old is that everyone, even people you gave birth to and raised, patronize you and handle you with kid gloves. Even though your best bet would have been to not spell the word, it's possible your grandmother appreciated having an adult conversation, if just for a little while.
She didn't care though and laughed about it when they told her what it was. She even mentioned something about women doing it quite often or something like that. She didn't care. Maybe you still consider it rude. Oh well.....
After rereading the opening post, I would say your only serious fault was not playing QUEENS or QUEERS instead. Granted, both of those words could cause offense, but it's a hell of a lot easier to explain them to family. It would only cost you a point as well.

I see how the sisters think it was rude, but honestly if everyone laughed about it and were not offended then it is no big deal. I know how sisters are though..
